National Technical Reports Library

The National Technical Information Service acquires, indexes, abstracts, and archives the largest collection of U.S. government-sponsored technical reports in existence. The NTRL offers online, free and open access to these authenticated government technical reports. Technical reports and documents in its repository may be available online for free either from the issuing federal agency, the U.S. Government Publishing Office’s Federal Digital System website, or through search engines.

Final Report on the Reproductive Toxicity of Formamide (FORM) (CAS No. 75-12-7) in CD-1 (Trade Name) Swiss Mice. Volume 1.

PB93109213

Publication Date 1992
Page Count 336
Abstract Formamide (FORM), a high production, industrial solvent with likely human exposure in industrial and laboratory settings, was evaluated for reproductive toxicity in CD-1 (Swiss) mice using the Reproductive Assessment by Continuous Breeding Protocol (RACB). Male and female mice were exposed to FORM in drinking water at doses of 0, 100, 350, and 750 ppm (approximately 20 to 200 mg/kg/day). A Maximum Tolerated Dose (MTD) of FORM, based on indices of generalized toxicity, was not reached for F0 animals. However, reproductive toxicity, including reduced fertility and litter size, was observed at 750 ppm FORM. F0 females at 750 ppm FORM exhibited an increase in the average days to litter, and tended to spend more time in diestrus than in any other stage of the cycle. F1 postnatal survival was unaffected by treatment. The MTD for the F1 generation was 750 ppm FORM, based on reduced body weight. Mild reproductive toxicity, observed as a reduction in litter size and increased days to litter, occurred at 750 ppm. No significant adverse histopathology was observed in either males or females. Estimated exposure to FORM was 26, 87, and 190 mg/kg/day for F1 animals.
