Acute oral toxicity was evaluated in 4 groups of 10 fasted and 4 groups of 10 non-fasted male Crl-CD rats administered single doses of bromobenzene by oral gavage at levels of 2600, 3000, 3500, and 3600 mg/kg of body weight (fasted) and 2900, 3400, 3700, and 4000 mg/kg of body weight (non-fasted). Mortality occured within 4 days of dosing and was observed for fasted animals in 8 rats at 3600 mg/kg, 6 at 3500 mg/kg, 7 at 3000 mg/kg, and 1 at 2600, while in the non-fasted animals, mortality was observed in 8 rats at 4000 mg/kg, 4 at 3700 mg/kg, 4 at 3400 mg/kg, and 1 at 2900 mg/kg of body weight. The LD50 was calculated to be 3068 mg/kg of body weight for the fasted animals and 3629 mg/kg of body weight for the non-fasted animals. Weight loss was noted for all animals at all levels. Clinical observations included weakness, stained face, stained and wet perineal area, chromodacryorrhea, moribundity, fasiculations, lacrimation, congestion, diarrhea, humped posture, tremors, convulsions, pallor, prostration, lethargy, and gasping. Gross necropsy was not evaluated. |
