Teratogenicity of test substance 5601-56-1 (Naphthalene) was evaluated in 5 groups of 4 New Zealand White rabbits dosed 0, 50, 250, 630, and 10000 mg/kg per day by gavage gestation days (GD) 6-18. Maternal weight gain was significantly reduced GD 12 in the 630 and 1000 mg/kg dose groups and GD 18 in the 630 mg/kg group. Food consumption decreased significantly in the 630 mg/kg group GD 7-10, 12-14, and 16-17 and in the 1000 mg/kg group GD 8-15. Maternal toxicity was shown by 50% mortality and 50% abortion rate at 630 mg/kg and 100% mortality at 1000 mg/kg. Dose related effects on dams were decreased body tone, loose stools, salivation, lacrimation, eye congestion, conjunctivitis, body drop, and cyanosis in 630 and 1000 mg/kg/day treated rabbits. On Cesarean section GD 29, total number of implantations, corpora lutea, viable and non-viable fetuses, and pre- or post-implantational losses (including early and late resorptions) were not significantly different from controls. Uterine weights were not reported. No fetal malformations or fetal deaths were found. Statistical significance was determined by one-way analysis of variance. |
